• Supplemental Figure 1 - GripTite^TM 293 cells were transfected with CaR/no PKC site plus pcDNA or different GRK 2 constructs and and subjected to IP assays 48-72 hours later. Normalized IP responses (%) were determined as IP formation in 10 mM Ca²⁺_e, minus IP formation in 0.5 mM Ca²⁺_e, normalized to the control condition (CaR plus pcDNA). Data of two independent transfections are shown below.
• Supplemental Figure 2 - Ca²⁺_e-dependent concentration response curves of CaR-mediated inositol phosphate (IP) formation as a function of β-arrestin expression (downregulation of both β-arrestins by transfection with specific siRNAs). Curves were fitted with GraphPad PRISM 3.02, using the following equation: Y = R_bas + (R_max-R_bas)/(1 + 10^((logEC₅₀-X)*n)).
