• Supplemental Figure - Supplemental Fig. Addition of pyruvate decreases the cellular level of NAD(P)H in 10 mM-Glc culture. N-38 neurons were initially seeded to Nunc 96-well plates and grown in 25mM-Glc media to be 70-80% confluent. Then they are switched to 0.5mM-Glc, 10mM-Glc media with/without 10mM lactate or pyruvate for 16 hours. Total cellular NAD(P)H level was measured on Twinkle LB 970 fluorometer (Berthold) by means of detection of autofluorescence of NAD(P)H (typical emission and excitation wavelengths are 440-470nm and 360-390nm, respectively). Data are calculated from fluorescence readings as relative percentages and presented as mean±SEM (n=4-6). Statistical difference was determined by Student's t-test (*p<0.05).
